When a plant is grown in the absence of light, the process that will be most affected is the light-dependent reactions.

The light-dependent reactions of photosynthesis directly require light to function. In these reactions, light energy is absorbed by chlorophyll and other pigments, which then drives the production of ATP and NADPH (energy carriers) needed for the subsequent Calvin cycle. Without light, these reactions cannot occur, so the plant will be unable to generate the energy it needs to drive the rest of the photosynthetic process.
